Yom Kippur (the Day of Atonement) is the holiest day of the Judaic Calendar, when we afflict ourselves (fast), pray, and seek forgiveness from our G-d and our fellows. It is the day when we are commanded to come closer to HaShem. It is the holiest day of the High Holy Days.
